The effect of fasudil on the activity of acetylcholinesterase in acute acephate poisoning rats |

Abstract Objective To study the effect of fasudil on the activity of acetylcholinesterase(AchE) after acute acephate poisoning and its possible mechanism. Methods A total of 72 male SD rats were randomly divided into 12 groups (n=6): normal saline group (group A), poisoning group (group B), conventional therapy group (in chronological order: C1, C6, C12, C24, C48group) and conventional therapy combined with fasudil group (in chronological order: F1, F6, F12, F24, F48group). The normal saline group was given 0.9% sodium chloride solution in the abdominal cavity. Each rat of the remaining 11 groups was given 30 mg/kg of acephate in the abdominal cavity to establish acute acephate pesticide poisoning models. The conventional treatment group was intramuscularly injected atropine (10 mg/kg) and resolve pralidoxime (20 mg/kg) after poisoning. The conventional therapy combined with fasudil group was intramuscularly injected atropine (10 mg/kg) and pralidoxime (20 mg/kg) and abdominally injected fasudil (10 mg/kg) for treatment. The complete blood and left ventricular tissue were taken at 1 h, 6 h, 12 h, 24 h and 48 h after poisoning. HE staining was used to observe the myocardial tissue injury. Serum AchE levels were measured by ELISA, and the expression of Bcl-2 and Bax were measured by Western blotting. Compare the changes in the measured indicators for each group. Results The survival rates of group F and group C were significantly higher than that of group B, and the survival rate of group F was significantly higher than that of group C. The difference was statistically significant (P<0.05). The levels of serum AchE in group C and group F were significantly higher than those in group B. The difference was statistically significant (P<0.05). Compared with group C, the level of serum AchE in group F was significantly higher than group C. In the conventional treatment group, the ratio of Bax/Bcl-2 in serum peaked at 12 h and the levels of acetylcholinesterase peaked at 6 h. In the fasudil combined with conventional treatment group, the ratio of Bax/Bcl-2 in serum peaked at 6 h and the levels of acetylcholinesterase peaked at 12 h. Conclusion Fasudil inhibits the activity of Rho kinase, relieves the inhibition of AchE in serum to a certain extent, and causes the peak of AchE level to migrate, thus reducing the damage of acetylcholine accumulation to myocardium.
