| Infulence of Neostigmine on bungarotoxin induced masthenia in rats |

Abstract Objective: To investigate the effect of Neostigmine on myasthenia of rats which is poisoned by bungarotoxine at early stage. Methods: SD rats were randomly divided into eight groups. A group was neostigmine 0.09 mg/kg used 15 min after poisoned. B group was neostigmine 0.09 mg/kg used 30 min after poisoned. C group was neostigmine 0.09 mg/kg used 1.5 h after poisoned. D group was neostigmine 0.18 mg/kg used 15 min after poisoned. E group was neostigmine 0.18 mg/kg used 30 min after poisoned. F group was neostigmine 0.18 mg/kg used 1.5 h after poisoned. G group was untreated after poisoned. H group was blank group. The rats were given celiac anaesthesia with 10% chloral hydrate 0.03 mL/100 g, and then fixed to mounting plate to take repetitive nerve stimulation check. After given injected subcutaneously with bungarotoxin 0.24 mg/kg, the EMG attenuation values, and partial pressure of oxygen were recorded. All of the index was statistical analyzed. The diaphragm, gastrocnemius were stained with HE. Results: There was no statistically significant difference in attenuation values, and partial pressure of oxygen between the groups treated with neostigmine and the control group(P＞0.05). There was no statistically significant difference in attenuation values, and partial pressure of oxygen between the groups treated with the same dose of neostigmine at different time(P＞0.05). There was no statistically significant difference in attenuation values, and partial pressure of oxygen between the groups treated with the different dose of neostigmine at the same time(P＞0.05). Pathological examination show the cells of diaphragm and gastrocnemius muscle swelling, nuclear disorganized at high magnification. There was no statistically significant difference between the groups. Conclusion: The effect of neostigmine to antagonize muscle weakness caused by bungarotoxin in poisoned rats is not observed.
