Abstract Poly(ADP-ribose)polymerase(PARP)plays a vital role in DNA repair and genome stability in eukaryotic cells. Over-activation of PARP is involved in the pathophysiological processes of gene regulation, signal transduction and cell apoptosis, and plays an important role in the process of acute inflammation. This paper reviews the structure and biological function of PARP, and study progress of PARP as a therapeutic target in several acute severe diseases such as ischemia-reperfusion injury,multiple organ failure, shock, and acute pancreatitis. Besides, it also summarizes the roles and possible mechanisms of PARP and its inhibitors in acute critical illness.
