Protective effects of ulinastatin on blemycin A5-induced pulmonary fibrosis in rats |

Abstract Objective To investigate the therapeutic effect and mechanism of ulinastatin on blemycin A5-induced pulmonary fibrosis in rats. Methods 75 rats were divided into normal group, model group and ulinastatin group. Rat pulmonary fibrosis was induced by intrabronchial injection of blemycin A5(5 mg/kg). Treatment was started from day 2 to day 28 with ulinastatin(20 000 U/kg). The pathological changes were observed by hematoxylin and eosin(HE) staining and expressions of transforming growth factor-β1(TGF-β1) protein and tumor necrosis factor-α(TNF-α) protein were tested by immunohistochemical technique. Results The pulmonary alveolitis and fibrosis were alleviated remarkable in the lung pathological section in ulinastatin group compared with those in the model group(P<0.01). The expression of TGF-β1 protein and TNF-α protein were decreased significantly than those in the model group(P<0.01). Conclusion Ulinastatin ameliorates the pulmonary fibrosis and its mechanisms may be through decreasing the expression levels of TGF-β1 protein and TNF-α protein.
