Effects of rhubarb extract on the expression of NSE and S100β in the brain tissue of rabbits after cardiopulmonary resuscitation |

Abstract Objective To study the preventive effects of rhubarb extract on the brain injury after cardiopulmonary resuscitation (CPR).Methods 24 male Japanese white rabbits were randomly divided into control group, CPR model group and Rhubarb group, 8 rabbits in each group. Rhubarb group were treated with Rhubarb extract 5 mL/(kg·d); control group and CPR model group were fed with 0.9% saline 10 mL/d. After 1 week, rabbits of CPR model group and Rhubarb group were produced ventricular fibrillation (VF) induced by 50 V alternating current stimulation to prepare CPR model. The rabbits of the CPR model group and Rhubarb group were sacrificed 2 hours after successful resuscitation, and the animals in the control group were sacrificed after anesthesia. The expression of NSE and S100β in brain were examined by immunohistochemistry.Results Compared with the control group, the positive expression of NSE and S100β was significantly increased in the CPR model group and Rhubarb group [NSE(n): 10.26±2.58, 8.80±2.66 vs. 5.39±1.47, S100β (n): 8.65±2.51, 6.73±1.74vs.4.58±1.87, all P<0.05]. Compared with the CPR model group, the positive expression of NSE and S100β were significantly decreased in the Rhubarb group [NSE (n): 10.26±2.5 vs. 88.80±2.66, S100β (n): 8.65±2.51 vs. 6.73±1.74, all P<0.05].Conclusion Cardiac arrest (CA) can lead to brain injury. Rhubarb extract can reduce the expression of NSE and S100β in brain tissue of rabbits after CPR, and protect the brain injury after CPR.
