Effect of NLRP3/caspase- 1 pathway in paraquat- induced acute lung injury |

Abstract Objective To investigate the effect of NLRP3/caspase-1 pathway in acute lung injury induced by paraquat (PQ) poisoning. Methods Thirty-six male Sprague-Dawley rats were equally and randomly assigned to two groups: PQ poisoning group (paraquat 20 mg/kg, ip) and control group (saline 1 mL,ip). Serum and lung tissues were collected at 8 h, 1 d and 3 d after PQ administration. The concentrations of
IL- 1 β and IL- 18 in the bronchoalveolar lavage fluid (BALF) were measured via ELISA assays. The contents of MPO and MDA in rats lungs were detected via colorimetric method. The mRNA and protein expression of NLRP3, ASC and caspase- 1 in lung were also determined via realtime PCR and Western blot, respectively. Location analysis of NLRP3 in lungs tissue was conducted via immunohistochemistry. Results Compared to the control group, the increased levels of cytokines including IL-1β and IL-18 in BAFL and the increased contents of MPO and MDA in lungs of PQ-treated rats were observed, as well as the mRNA and protein expression of NLRP3, ASC and caspase-1 in lungs. The NLRP3 protein was slightly expressed in normal lung tissues and increased three days after PQ administration. Conclusion NLRP3/caspase- 1 pathway is activated in paraquat-induced acute lung injury, and then the inflammatory cytokines IL-1β and IL-18 expression and secretion are caused subsequently.
