Vitamin D3 is involved in the regulation of pulmonary dendritic cell activity in septic rats |

Abstract Objective: To understand the mechanism of vitamin D3(Vit D3) in the treatment of sepsis through the regulation of pulmonary dendritic cells (DCs), we examined CD83 and CD86 and anti-inflammation and proinflammatory cytokines in septic rat model. Methods: ①Female juvenile rats (n=24) were randomly divided into three groups including normal control group, septic group and Vit D3 intervention group. ②The rats in the septic group and Vit D3 intervention group were intraperitoneal injected with lipopolysaccharide (LPS) for preparation of septic model. ③The expression of CD83 and CD86 on the surface of pulmonary DCs in the rats was detected using flow cytometry. ④Proinflammatory cytokines (IL-12) and anti-inflammatory factor (IL-10) in the lung tissue of rats and 25 (OH) D3 in the rat serum were detected by ELISA. Results: The expression levels of CD83 and CD86 on the surface of pulmonary DCs in the septic group (6.81±0.44)% and( 5.52±0.67)% and Vit D3 intervention group (4.58±0.47)% and (3.19±0.50)% were significantly increased, comparing to the normal control group (0.86±0.13)% and (0.54±0.16)% (t=36.115,18.128 and 18.735,18.908,P<0.05), but the expression levels of CD83 and CD86 in the Vit D3 intervention group were lower than those in the septic group (t=13.326,7.433,P<0.05), respectively. The inflammatory factor IL-12 in the lung tissue in the Vit D3 intervention group (114.71±45.61) ng/L was reduced comparing to the septic group (343.86±97.87)ng/L(t=8.595,P<0.05), whereas the anti-inflammatory factor IL-10 level in the Vit D3 intervention group (194.18±24.53) ng/L was higher than that in the septic group (150.59±32.48) ng/L(t=2.933,P<0.05). The serum 25(OH) D3 level in the Vit D3 intervention group (41.76±7.97) mg/L was increased compared with the septic group (21.58±8.04) mg/L(t=6.941,P<0.05), but serum 25(OH) D3 level in these two groups were significantly lower than that in the normal control group (60.24±5.67)mg/L(t=8.388,10.050,P<0.05). Conclusion:Vit D3 may inhibit the activity of pulmonary DCs in septic rats and alleviate the excessive immune response and inflammatory response in sepsis.
