Expression ofintercellular cell adhesion molecule - 1mRNA in Sprague Dawley rats with transfusion - related acute lung injury |

Abstract Objective To study the expression of intercellular cell adhesion molecule - 1 (ICAM - 1)and the number of polymorphonuclear neutrophil( PMN) in rats with transfusion - related acute lung injury,to explore their possible role in TRALI. Methods Sixty Sprague Dawley( SD) rats were rando mLy divided into normal control group (n = 15) with sham operation, positive control group (n = 15) with ALI induced by intravenous infusion of lipopolysaccharide ( LPS) ( 5 mg / kg), and TRALI group (n = 15) treated by intraperitoneal injection of LPS 2 h before the transfusion of human plasma(about 1 mL), LPS control group (n = 15) treated by intraperitoneal injection of LPS 2 h. The reverse transcription - polymerase chain ( RT - PCR ) was used to detect ICAM - 1 mRNA. Andimmunohistochemical method was used to detect ICAM - 1 protein expression,Morphological changes in lung was observed under light microscope. Myeloperoxidase (MPO) in lung homogenate,wet lung weight to dry lung weight ratio(W / D) were observed. Results Compared with normal control group and LPS control group, Expression of ICAM - 1protein in plasma were increased significantly in positive control group and TRALI group(0. 58 ± 0. 09、0. 41 ± 0. 07 vs. 0. 12 ± 0. 03、0. 21 ± 0. 04) ( all P < 0. 01); Compared with normal control group (0. 16 ± 0. 03) and (0. 36 ± 0. 05),positive control group(0. 33 ± 0. 09 and 1. 53 ± 0. 17) and TRALI group (0. 27 ± 0. 07 and 1. 24 ± 0. 11),Expression of ICAM - 1 protein and ICAM - 1 mRNA were increased significantly in the lung ( all P < 0. 05); TRALI group [(310 ± 76. 6) × 106 / L and (33. 5 ± 11. 5)% ],the number of cells in BALF and PMN percentage were significantly higher than that of normal control group[(101 ± 63. 8) × 106 / L and (9. 8 ± 3. 5)% ] (all P < 0. 05); TRALI group, tissue water content and MPO activity is significantly higher than normal control group(all P < 0. 05). Conclusion Expression of ICAM - 1 protein and ICAM - 1 mRNA were increased in the rat lung with TRALI and PMN infiltration in lung tissue,suggests ICAM - 1 participate in the process of the PMN and endothelial cell adhesion,indicating ICAM - 1 and PMN may play an important role in the TRALI of the inflammatory.
