Abstract Objective To investigate the effects of Berberine(BBR)on lipopolysaccharide(LPS)-induced damage to human umbilical vein endothelial cells(HUVECs)in vitro. Methods The endothelial cells were cultured in vitro. HUVECs were pretreated with various concentrations BBR(1.25,2.5,5.0 μmol/L)for 24 h before exposure to 5 μg/mL LPS for another 24 h. The ability of cell proliferation was detected by EDU incorporation assay. The cell cycle distributions and apoptosis were analyzed by flow cytometry. The protein levels of phospho-c-jun N-terminal kinase(p-JNK)were detected by Western blot. Results LPS treatment significantly decreased HUVECs viability,decreased the G2/M phase cells,increased the apoptosis rate and increased the phosphorylation level of JNK. Pretreatment with BBR could increase cellular viability,increased the G2/M phase cells,decreased apoptosis rate and reduced the level of p- JNK in a dose- dependent manner. Conclusion BBR can effectively reduce LPS-induced damage to HUVECs by inhibiting apoptosis and down-regulating the phosphorylation of JNK.
